DSPE-PEG-Amine to DSPE-PEG-SH

Task: Reconstitution of DSPE-PEG-Amine into Chloroform
- Added 200 uL of chloroform into amber DSPE-PEG-Amine vial (from Avanti), mixed and transferred to glass vial via pipetting, repeated with another 200 uL volume and a 100 uL volume for a total of 500 uL chloroform
- After reconstitution, attempted to add 200 uL and 300 uL aliquots to microcentrifuge tubes, however noticed there was a volume loss
- To ensure volume loss came from evaporation and not spillage we evaporated the rest of the chloroform and weighed the tube, comparing to the weight of the tube prior to addition. Noticed that there was only 4.1 mg remaining
- Decided to order more DSPE-PEG-Amine, will use the remaining 4.1 mg for H-NMR and thiol assays, remaining DSPE-PEG-Amine stored in the -20
Task: Reconstitution of 25 mg Traut’s regent into 3 mL methanol
- Made 3 aliquots of reconstituted Traut’s reagent. This is stored in 4 degree fridge

Task: Thiolate DSPE-PEG-Amine
- Added 15 uL of stock Traut’s reagent to DSPE PEG Amine
- Added 95 uL methanol
- Left to dry into powder – will start dialysis tomorrow
Trial 2 with new DSPE-PEG-Amine

Task: Synthesis of DSPE-PEG-SH
- Decided to use all 25 mg since it’s an easier amount to work with
- Dissolved 25 mg of DSPE-PEG-Amine in amber vial with 500 uL chloroform
- Added 75 uL Traut’s stock
- Added 475 uL Methanol
- Left out to dry overnight in the fume hood
